This week Dr. Karina Rios II Graduate Resident of Dermatology, Institute of Biomedicine, Central University of Venezuela, has an article entitled "Topical Antihistamines express a potent download anti-inflammatory activity linked in part to the improvement in barrier permeability" published in the Journal of Investigative Dermatology 2013, 133:469-478. The authors are Lin TK, Man MQ, Santiago JL, Park K, Roelandt T, Oda Y, Hupe M, Crumrine D, Lee HJ, Gschwandtner M, Thyssen JP, Trullas C, Tschachler E, Feingold KR, Elias PM.
Systemic histamine antagonists, the type 1 and type 2 receptors (H1/2r) are widely used as anti-pruritic and sedative, but demonstrated a modest anti-inflammatory activity.
Since many inflammatory skin diseases are the result of defects in the skin barrier function, and that keratinocytes produce Hr1 and Hr2, researchers hypothesized that topical application of H1/2r antagonists should be effective in treating inflammatory dermatoses.
Topical application of antagonists H1/2r restoring improved permeability of the skin of normal mice by the following mechanisms: (i) stimulation of epidermal differentiation, which leads to thickening of the cornified pockets, and (ii) increased synthesis and secretion of epidermal download lipids.
As barrier homeostasis download was significantly improved in mice deficient in mast cells, the topical application of agonists H1/2r produced a further improvement, which follows her that the synergistic download effect of the antagonists act by different ways, but its main target to the mast cells.
